How to Proceed:

Step 5: Press the "Vertical Import" button if you want to import your date in a vertical manner (A1, B1, C1, etc.) or "Horizontal Import" in case you prefer a horizontal arrangement of your data on the plate (A1, A2, A3, etc.)

Premixed Primer:
Primer added by customer.

Standard Primer List:
Primer added by Microsynth. Select your sequencing primer from a list of 94 standard primers which are stored at Microsynth (primarily for plasmids).

Enclosed in Separate Tube:
Primer added by Microsynth. Customer ships primer and DNA in separate tubes to Microsynth.

Stored at Microsynth:
Primer added by Microsynth. Prerequisite for this option: You have already sent the primer to Microsynth in the past. Your primer will be stored for 6 months at Microsynth. The primer name will be automatically deleted from this list after 6 months or if the primer has been used up.

Custom Primer List:
Primer added by Microsynth. This option is an add-on to the option "Stored at Microsynth". If you know that a primer will be used frequently in future orders, send us a larger volume of this primer (at least 50μl) and add it to your custom primer list. We will then store it for an extended period (up to one year).

Order Now:
Primer added by Microsynth. Via this option you can order a new primer already during the order process without changing the order form. While your DNA is on the way to Microsynth, we will produce your sequencing primer and forward it to our sequencing lab.

Design at Microsynth:
Primer added by Microsynth. This option is similar to "Order Now". In addition to primer synthesis Microsynth performs also the primer design. You just have to provide us by email with a sequence of 50 - 200 bases.

Please choose this option in case you want Microsynth to purify your PCR product (removal of PCR primers and dNTPs). Please note that this service is only available at an additional charge.
Please mark this checkbox only if you know that your sample contains GC-rich sequences or hairpins.
Please choose this option only in case you have difficult-to-sequence samples which have not been successfully sequenced by using our standard sequencing protocols (e.g. GC-rich Sequence).

Please note that the “Advanced Treatment” protocol is subject to following limitations/conditions:
- all samples of an individual order have to be marked with this specific sequencing protocol
- is not compatible with overnight sequencing
- is only available at an additional charge

If you work primarily with specific sequencing primers which you will use again and again over the coming months, Microsynth recommends you to make use of the “Custom Primer List”.

How does it work?
- If you have choosen the primer source „Order Now“ or „Enclosed in Separate Tube“, simply mark this box and the specific primer(s) of this order will be added to your „Custom Primer List“
- Or alternatively go to the “Options & Preferences” section and select “Review/Edit Custom Primer List” under “Primer Management”. Here you can add and delete specific sequencing primers.


Your overall benefits:
- Your specific primers will be stored at Microsynth
- When placing your next order, you can easily select the requested primer(s) from your “Custom Primer List” by one simple click.
- When working in a larger lab, you will have the possibility to share specific sequencing primers amongst your research group. For this purpose, however, you have to first create and define your group. This can be done within the “Options & Preferences” section when selecting “Create Group/Show Group Members” under “Group Management”.
Our Special Sequencing Protocols are especially useful if you want to decode difficult-to-sequence constructs such as regions containing hairpin structures, G/C-rich sequences, etc.
Continuous optimization of our Sanger sequencing capabilities has resulted in protocols that are able to map difficult or even very difficult DNA templates with a success rate >98%.
Generally, we recommend the following two-stage approach to our customers:
Single Strand Sequencing (Accuracy >99,90%) Double Strand Sequencing (Accuracy >99,999%).
With this pre-test, a few samples of your 96-well plate are picked out and sequenced. If the pre-test shows that quality/concentration of your DNA template and/or your primer are in the appropriate range, your entire plate will be sequenced. This initial test phase is free of charge but will delay delivery of your sequencing results by one working day. Microsynth generally recommends this pre-test unless you know in advance that requirements are given for yielding good sequencing results.

The primer is not stored at Microsynth.
Possible reasons:
  1. The spelling of the primer name is wrong.
  2. The expiry date has exceeded and the primer was discarded.
  3. The primer has never been stored at Microsynth. Please check if you entered the correct primer source.
  4. The primer name is not yet saved in our sequencing data base because you plan to order the primer in the oligo webshop after this sequencing order. In this case, please choose the primer source “Design at Microsynth” or "Order Now".
